    After completing masters my brother changed from F1 to H1B visa.
    After one year due to recession layoff, he shifted from H1B to F1 again (second MS).
    Nearly 6 months later he got a new job and so he discontinued his second MS and changed to H1B again.
    He hasn't done visa stamping yet.
    He is planning to visit India shortly (after one year of changing to H1B visa).
    This'll be his visit to India after 3.5 years.
    Will he face any problems in visa stamping?
    Will he be able to return back to US without any problem?

    Applying a COS from H1B to F1 and back to H1B is not an issue. But whether he will get his H1B stamped or not is nobody's guess. It depends on what documents he carries and how well he takes the visa interview. If there was question about why he discontinued his 2nd masters, he should be in a position to justify that.
