H1 transfer from Non Profit to For Profit

Hi, I am on Non Profit H1. I had a For profit H1 before 6 years. And I am going to transfer my H1 to For profit again.
My question is , when should I start working with my new employer ? After USCIS receipt OR after H1 transfer from Non profit to For profit?

Are you saying that your H1 tenure has completed 6 Years? Doesnt matter for profit or non profit H1.
