I didnt travel to US after getting my H1B in Oct2007 which is valid upto Sep 2010.Im planning to travel now.Can I travel now and if I travel what will be the questions raised in Port of Entry?

Yes, you can travel as long as the visa is valid. There will be no special questions because of your delayed travel. All that the officer at the POE will be interested in is your project related information (So carry the latest employment authorization letter). It is not necessary to travel immediately after the visa is approved.
