My mother and sister are going to apply for B2 visa in few days. Will they need to write separate HDFC bank cheques? If yes, then which receipt number they will need to use to take an appointment?

Yes, each needs a separate payment and will get a receipt number for using on the application to book the appointment online.

Okay. But which receipt number will they need to use to take an appointment? Receipt number given to my mom or the number given to my sis?

Each gets one receipt number. First start filling out the application for one person using the receipt number, when done, it will ask if you want to add additional people. The receipt number of the second person has to be entered at that time.
