You need a Canadian Visa before any trip there, I believe because Canada and USA are two different countries! You have USA visa, not Canadian, correct? And as long as you have a visa ( Canadian not USA) to enter Canada, it should not be a problem: just don't overstay!
About the I-94, I don't have clue what the immigration is going to do, but one thing is sure they will ask you bunch of questions about this trip!

      1) depending of your country of citizenship, you may need a visa to enter Canada
      2) if you do need a visa for Canada - the canadian embassy/consulate where you apply may (or not) frown at the fact you're applying for it here, and not in your home country. If they grant you a visa, (3) applies
      3) they visa will have an expiration date of either (a) your passport expiration date, or (b) your I-94 date

And no - if you DO travel to Canada, they won't take your I-94

And no - traveling to Canada while visiting the US shouldn't have any effect on your next visit to the USA

Though: depending on frequency of your visits to the US, and your length of stay on each visit, the CBP agent will adjust your allowed stay in the US. In other words - if you enter the US on 1/1, you will probably get a six-month stay. If you actually stay for a couple weeks, and you come back on 2/2, you will probably again get a six-month stay. If, on the other side, you arrive on 1/1, get a six-month stay, and leave on 6/29 - the next time you enter the US, you will probably get a *way shorter* allowed stay - and if you keep coming too frequently and staying for too long, you may even be denied entry.

In other words: coming every two or three months and staying for a week each time is within the pattern of usage expected for a B1/B2 visa - you may be coming for business meetings, training, attending a conference, you name it. Coming for six months, leaving for a week, and coming back for "another six months" is not.
